Typically the only thing that would produce additional lettering on a coin that is also out of place would be a die clash. I checked the site Mad Die Clashes for the overlay of a Jefferson nickel and found that there would be no image of a letter in the space where you believe to be an extra letter on your nickel. Therefore, I agree with Greenstang that it just appears to be an N but it is just circulation and the way the coin has developed its coloration from circulation.

Why all the secrecy concerning the date of issuance of the coin?

And why the use of highly radioactive charged plutonium rods to prop the coin up?
